Of Wellfleet. He and his brother Elisha married two daughters of Daniel Mayo Sr. of Eastham, whose will (1754) names "Martha the wife of Ezekiel Holbrook." The Eastham record reads: "Ezekel holbrooks and martha mayo boath of Eastham ware marrid in Eastham by Mr Isaiah Lues minestar in octobar 10 : 1751." Their children are recorded there: Sarah 1752, Mercy 1753, Martha 1757 and Samuel 1759. Martha died 7 June 1775.

In 1765 he was one of the men who set off the widow's thirds in the estate of Thomas Holbrook late of Eastham, probably his brother.
